 I'm trying to figure out the least stressful way to get my horses from the NW side of San Antonio to Bryan, TX....

One of Mapquests options was to go through Dripping Springs on Rt. 290 and then on TX 71 to Bastrop....  The question is how busy is Rt 71 on a Tuesday mid-morning around Austin...?  Any major constructions or other drama?

I have tried all the other ways to get over there and they are all not good.  Any insight on this road would be appreciated...

 I think if you can time it so that you are coming thru Dripping Springs/Austin between 9am and 11am you should be OK.  Most of the work traffic should be thru by then.

 ETA:  I live east of Austin.  When I come down to the San Antonio Rose Palace I take the 130/45 Toll Road from Manor down to Buda and then I35 down to the Loop 1604 in San Antonio.  That works pretty well for me.

I live just east of Austin and would just use I35 and you could either go through Bastrop or take the toll to 290. Mid morning you should be fine. They have completed the fly over at I35/71 so traffic isn't as awful to take I35 all the way up if you didn't want the toll fees. 
ETA: The only cruddy thing about going through Dripping Springs is that there are a lot of stop lights on 71 to Austin (and the drivers are typical "city" drivers that are not as trailer friendly), not as many lights from Austin to Bastrop, but there are some. If you're not wanting to make a lot of stops I would just take the toll to 290 and you should be able to roll right through Manor & Elgin.
